Manual nucleic acid extraction is a lab technique for isolating DNA and RNA without automated machinery. It involves four main steps: lysis, where a buffer breaks open cells to release nucleic acids; separation, often using a phenol-chloroform mix to separate the nucleic acids from other cellular components; precipitation, where alcohol and salt are added to make the DNA/RNA form a pellet; and finally, washing and elution, where the pellet is cleaned and re-dissolved in a buffer for future use. This labor-intensive method yields highly pure nucleic acids.
